Parainfluenza viruses serotypes 1, 2 and 3 cause laringotracheobronchitis (croup) in children between two and four years of age. The serotype 3 is the second agent producing pneumonia in children under one year (after respiratory syncytial virus) and it has an epidemic pattern. Serotypes 1 and 2 are frequently associated with croup and have and endemic pattern. Current methods for serodiagnosis are virus neutralization, hemagglutination inhibition, indirect immunofluorescence, complement fixation and ELISA tests. ELISA is a sensitive and specific test. ELISA IgM tests are specially indicated in infants less than 6 months, in order to avoid interference by maternal IgG. Besides, a rapid diagnosis may be achieved by the detection of IgM antibodies in a serum drawn at the onset of the disease. Detection methods based on chemiluminescence have received much attention due to their low background, linearity and wide dynamic range. When coupled to enzyme immunoassays, the signal amplification effect provided by the enzyme enables the design of CLIA (ChemiLuminescent ImmunoAssay) tests with shorter incubation times while keeping or improving their sensitivity.
